Here how I use Aqua Pens:
Filling the Pen: Unscrew the water container (clockwise to open) and fill with water. Reattach and squeeze to wet the brush tip. These pens screw on "lefty-tighty, righty-loosey," making them more secure against leaks and preventing accidental opening.
Blending Colors: Apply color directly with the pen and apply it to the paper. This pen can be also used for activating watercolor pencils, or working with gouache and ink.
Controlling Water Flow: Squeeze the barrel gently for less water and harder for more flow. For fine lines, use light pressure; for wider strokes, use the side of the brush.
Cleaning the Tip: Squeeze the barrel to push water through the brush and rub it gently on a paper towel or in a water container.

Side note: The following tools are all commonly referred to as "watercolor pens," though they serve different functions:
